Impact of clonal haematopoiesis with indeterminate potential mutations on transcription in single-cells (WP1)
Rationale and objectives
We need to understand how early mutations in clonal haematopoiesis with indeterminate potential (CHIP) provide a growth advantage and contribute to the emergence of myelodysplastic syndromes (MDS).
Therefore, we plan:
• To test and develop computational methods for the integration mutation status and gene expression of single cells.
• To determine specific transcriptional profile induced by selected mutations in CHIP and MDS on the single-cell level.
• To validate regulated genes in large cohorts of bulk and single cell data.
Envisioned secondments
Erasmus Medical Centre and/or Universitätsklinikum Aachen AöR (The Netherlands and/or Germany, 3 months), MLL Munich Leukemia Laboratory (Germany, 1 month).
